Abstract:
This project assesses the physiological status of selected macroalgae in Hawaii under various nutrient regimes, with the purpose of determining a suitable physiological test that can, with reasonable accuracy, define nutrient concentrations in the adjacent water column. The project is designed to test the utility of pulse amplitude modulated (PAM) fluorescence as a means of assessing physiological status. PAM fluorescence provides an estimate of photosynthetic efficiency and can be measured underwater in the field. This technique may be suitable for resource managers seeking to rapidly assess nutrient concentrations in the marine and aquatic environment. The data were taken from July 2001 - July 2002.

The chlorophyte macroalgae Dictyosphaeria cavernosa was collected at Coconut Island in Kaneohe Bay, Oahu, Hawaii,and acclimated for one week in flowing seawater at the Hawaii Institute of Marine Biology research station prior to deployment in the field. The algae were cleaned, weighed and deployed at 3-4 m depth at three patch reefs in KaneoheBay (North, Central and South Bay sites). At each site four treatments were set up, each with three replicate algal samples. Each treatment was positioned 10 m apart to minimise interference. The treatments were 1) no nutrient addition, 2) Nitrogen addition,3) Phosphorus addition, and 4) Nitrogen and Phosphorus addition. After one week in the field, light response curves were carried out in situ on algal samples early in the morning. Samples were removed in the dark and analysed for chlorophyll a and b content, CNP composition, 15N:14N and13C:12C ratios, and mass increase (growth). This was carried out monthly for 13 months. Data collection - Data are presented raw, except for light response curve data, of which final calculated parameter estimates are provided. Data analysis - Values of the light response curve parameters ETRmax and alpha were derived by non-linear curve-fitting techniques.
